No issue with running NT01s on the street. I have done it, albeit not for a 100 miles.
Stuff to consider though:
They absolutely suck in the wet, so if it rains, you're in trouble...
R-comps are more prone to pick up road debris and and as such are more prone to get a puncture. So consider the risk.
NT01 don't really need to be heat cycled or broken in.
